・Standard: The standup (rise up) of the colony was good for the second week. And, as for the colony, Rough type was conspicuous in the fourth week, and showed the form of peculiar M.tuberculosis, to dry, friable and irregular margin. However, dryness of a culture medium is conspicuous after the fifth week, and it seems that the colony stopped their growth.
・Test1: Early growth is slow, but it shows a strong positive at the fifth week. However there are few Rough types.
・Test2: Generally growth is good, Rough type is conspicuous and good growth (vigor is in growth) rather as compared with other test tubes from the fifth week.
・Test3: It seems that many amount of samples comparing others the 2nd week carrying out cultivation 2(+), although it almost comes out, in Flat, a colony is spiritless and is dysgonic growth.
Cause of poor growth in a long test tube
1] The supply state of oxygen was not good.
2] Since test-tube glass is thin, it is easy to be influenced of the temperature change by electric power failure.
3] Insufficient of the amount of inoculum by swab.
⇒With the long test tube, since the branch of swab was short, inoculation has not fully been taken in.
Recommendation:
I. Installation of the generator to incubator
II. To change a long test tube
III. Perform inoculate by swab.
IV. A pretreatment is shortened in 5 minutes.
