identical to those of B. subtilis (4.2 Mb and 43.7 mol%). The nucleotide sequence data of the B. subtilis chromosome will definitely bring us whole bunch of information to figure out common function in bacilli and will be a good model to analyze the C- 125 chromosome. From these research background, we have just initiated the genome analysis of C-125 based on the information of B. subtilis. As the first step of genome analysis, we attempted to complete a physical map for the C-125 chromosomc. The chromosomal DNA was digested into 20 fragments with AscI and almost all linker clones were isolated. We also attempted to construct lambda phage library of the C- 125 chromosome and sequence several clones. The nucleotide sequence data indicated that some gene clusters on the B. subtilis chromosome were rendomly conserved on the C-125 chromosome.
